A porous Cu(II)-MOF with one-dimensional hexagonal channels for solvent-free cyanosilylation and anti-gastric cancer activity by trigger ROS induced cell apoptosis
In continuation of our research interest in the construction of multifunctional metal organic frameworks (MOFs), we report here a novel porous Cu(II) MOF with the chemical formula of {[(Cu(L)(2)(H2O)(2)](DMF)(3))(n) (namely 1 hereafter) which can be synthesized by the self-assembly of 2-methyl-4-(1H-pyrazol-4-yl)pyridine (namely HL in the following texts) and Cu(II) ion in a mixed solvent of DMF and water under room temperature. The obtained MOF 1 has been characterized and analyzed in detail by single crystal X-ray diffraction, powder X-ray diffraction (PXRD), elemental analysis, Fourier-transform infrared spectra (FT-IR), and thermogravimetric analysis (TGA). Furthermore, in view of its water occupied open Cu(II) metal sites along with the uncoordinated N atoms as Lewis basic sites, the activated MOF 1 (denoted as 1a hereafter) can be applied as a heterogeneous catalysis for solvent-free cyanosilylation with high efficiency and size selectivity, and its possible reaction mechanism has also been proposed. After the design and synthesis of compound, the anti-cancer activity of compound on gastric cancer was further explored. Firstly, the CCK-8 assay was detected to determine the inhibitory effect of compound on cancer cell viability. Then, the Annexin V-FITC/PI assay and the H2DCF-DA assay was performed to measure the influence of compound on cancer cell apoptosis and ROS accumulation.
